Enzyme Kinetics - Effect of pH

Instructions:

  1. Select values of ETot and [S] that will give absorbances values approximately equal to 1.
  2. Keeping the enzyme concentration and substrate fixed, vary pH and measure product formation, use pH values from 2 to 9.
  3. Use an extinction coefficient of 18,300 M-1cm-1 to convert the absorption to product.
  4. Plot the rate of product produced/sec versus pH.

Note that your instrument cannot read absorbance values greater than 2.
